Ph.D. Thesis Description

Embryos of oviparous aquatic organisms are continuously exposed to water pollutants. A wide range of xenobiotics can damage the DNA of developing embryos, disturbing embryogenesis. My PhD. research work has evidenced that fish embryos have evolved a developmental stage-specific DNA repair machinery to combat DNA damage. This information is crucial for the ecological risk assessment of fish embryos. Additionally, several studies have reported the indispensable role of DNA repair proteins in zebrafish organogenesis using gene-editing approaches. However, under natural conditions, there can be either an increase or a decrease in protein expression in response to stress. Thus, we aimed to investigate how exogenous DNA damage during embryogenesis can disturb the basal expression of DNA repair proteins and how this disturbance impairs their developmental role. This can explain one of the reasons for embryonic malformations in response to genotoxic stress. We also characterized the species-specific DNA damage response during fish embryo development and identified that early-stage embryos are more sensitive to DNA damage than later stages. During my Ph.D., I worked on the evolutionarily conserved non-model species sterlet (Acipenser ruthenus), common carp (Cyprinus carpio), and zebrafish (Danio rerio). We chose two evolutionarily distant fish species to delineate the evolutionary conservation of DNA repair machinery during fish embryo development.
